Target Information
Cyclin D1 (PRAD-1, bcl-1)는 세포 주기 조절의 핵심 단백질로, cdk4 및 cdk6과 결합하여 Rb 단백질을 인산화함으로써 세포 주기 진행을 조절합니다. 이 단백질은 다양한 인간 종양, 특히 mantle cell lymphoma (MCL)에서 과발현되는 원암유전자(proto-oncogene)로 알려져 있습니다. Cyclin D1은 G1 phase 동안 합성되어 CDK4 또는 CDK6과 복합체를 형성하며, 성장 인자 자극에 반응하여 세포주기 G1/S 전환을 조절합니다. 인산화에 의해 Cyclin D1이 불안정해지고 ubiquitin 매개 분해를 통해 G1/S phase 억제에 관여합니다. Cyclin D1은 CDK 억제 단백질 p21Cip1 및 p27Kip1을 조절하며, 다양한 종양 발생 과정과 밀접하게 연관되어 있습니다.
