Target Information

Estrogen Receptors (ER) are part of the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor acts as a ligand-activated transcription factor; when bound to estrogen, it undergoes a conformational change allowing dimerization and binding to estrogen response elements (ERE) in DNA. This binding can either activate or repress gene transcription through interaction with coactivator or corepressor proteins.

There are two main forms of the estrogen receptor, alpha and beta, encoded by separate genes (ESR1 and ESR2). Due to alternative RNA splicing, at least four isoforms of estrogen receptor-alpha exist (Isoforms 1: 66 kDa, 2: 53 kDa, 3: 47 kDa, 4: 35 kDa). Estrogen receptors are widely expressed across tissues and are crucial for sexual development, reproductive function, and bone metabolism. They are also involved in pathological processes such as breast cancer, endometrial cancer, and osteoporosis.
