Product Specific Information

The Michel-17 monoclonal antibody reacts with mouse CD83, a 45 kDa cell surface glycoprotein belonging to the immunoglobulin superfamily. CD83 is mainly expressed on mature dendritic cells and activated lymphocytes. Cross-linking of CD83 with Michel-17 does not induce activation signals. CD83 plays a role in T cell development through ligand interaction, with CD83 ligand mainly expressed on B220⁺ cells in mouse spleen.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on LPS-activated mouse splenocytes. Recommended ≤0.5 µg/test (in 100 µL volume). Cell number: 10⁵–10⁸ cells/test. Optimize titration for best performance.

Target Information

CD83 is a 40–45 kDa glycoprotein expressed on peripheral blood dendritic cells. Peripheral lymphocytes can express low levels after stimulation (e.g., Con A, PHA). In immunohistology, CD83 is strongly expressed by interdigitating reticulum cells and weakly in germinal center cells, also present in Langerhans cells of the skin.

The CD83 antigen is a 186-amino-acid single-chain glycoprotein, a member of the immunoglobulin superfamily with an extracellular V-type Ig-like domain, transmembrane region, and short cytoplasmic tail (~40 aa). It undergoes extensive post-translational glycosylation, doubling its molecular weight compared to the core protein.

CD83⁺ cells show a unique phenotype distinct from T, B, NK, or myelomonocytic cells, coexpressing high MHC class II and various lineage markers (CD2, CD5, CD40, CD78, CD13, CD33, CD36). CD83 dysfunction has been associated with plague and Rift Valley Fever.
