Target Information

Epithelial sodium channels are amiloride-sensitive members of the Degenerin/epithelial sodium channel (Deg/ENaC) superfamily of ion channels. These channels share structural organization with two short intracellular termini, two transmembrane segments, and a large extracellular loop containing a conserved cysteine-rich region.
ENaC isoforms (alpha, beta, gamma) are expressed in kidney, lung, and colon, playing essential roles in sodium and fluid balance. ENaC mediates aldosterone-dependent sodium reabsorption in the distal nephron, regulating blood pressure.
ENaC function is partly regulated through interaction with the CFTR chloride ion channel. Mutations in ENaC subunits can lead to hypertension (Liddle’s syndrome) or pseudohypoaldosteronism (PHA-1).
