Product Specific Information

Description:
The MEM-15 monoclonal antibody reacts with human CD14, a 53–55 kDa GPI-linked glycoprotein expressed on monocytes, macrophages, and some dendritic cells. Complexes of LPS and LBP bind with high affinity to monocytes via CD14.

Each product contains 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.

Applications Reported:
Validated for flow cytometric analysis.

Applications Tested:
Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells. Use at 5 µL (1.0 µg) per test in a final volume of 100 µL. Cell number may vary from 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample (regardless of the number of NovaFluors).
  • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates use Phiton™ technology based on nucleic acid dye structures engineered to minimize spectral spillover and spread.

Target Information

CD14 is a 55 kDa GPI-anchored glycoprotein expressed on monocytes, macrophages, and neutrophils. It functions as a lipopolysaccharide receptor and is found both membrane-bound and soluble in serum. CD14 interacts with LBP to bind LPS and plays a role in immune response modulation, apoptosis recognition, and inflammatory processes. Dysregulation is linked to infections such as Mycobacterium chelonae and respiratory conditions like croup.
