Product Specific Information

The monoclonal antibody NVG-2 reacts with mouse CD63, also known as Lysosomal-Associated Membrane Protein 3 (LAMP-3) or tetraspanin 30 (TSPN30), a member of the tetraspanin family of proteins characterized by four transmembrane domains.
CD63 is expressed on various cell types of hematopoietic lineage (granulocytes, B lymphocytes, platelets) and non-hematopoietic cells. It is found on the cell membrane, late endocytic vesicles, lysosomes, exosomes, and other specialized granules.
On the cell surface, CD63 interacts with various proteins forming tetraspanin-enriched microdomains (TEM). Its high expression on the membrane may indicate cell activation, thus CD63 serves as an activation marker for basophils, platelets, and other cells.

Applications Tested

This NVG-2 antibody was tested by intracellular staining followed by flow cytometric analysis of mouse resident peritoneal exudate cells using the Intracellular Fixation and Permeabilization Buffer Set (Product #88-8824-00) and the BestProtocols® Protocol A: two-step protocol for cytoplasmic intracellular proteins.
Use ≤0.25 µg per test (defined as antibody amount for 100 µL sample). Cell number may range from 10⁵ to 10⁸ cells/test. Optimize titration for best performance.
Excitation: 488–561 nm
Emission: 578 nm
Laser Compatibility: Blue, Green, Yellow-Green Laser


Target Information

CD63 (LAMP-3) is a glycoprotein of the tetraspanin family present in late endosomes, lysosomes, and secretory vesicles. It is also found on the plasma membrane following cell activation, making it a widely used basophil activation marker.
CD63 interacts with integrins, affects phagocytosis and migration, and participates in regulation of ion channels and trafficking. It also functions as a T-cell costimulatory molecule.
Expression of CD63 can help predict carcinoma prognosis. It is expressed on activated platelets, monocytes, macrophages, and weakly on granulocytes and lymphocytes.
Diseases associated with CD63 dysfunction include melanoma and Hermansky-Pudlak Syndrome.
