Product Specific Information
For reconstitution, add 100 µL distilled water to obtain a final antibody concentration of about 1 mg/mL.
For conjugation experiments, perform desalting using Zeba Spin Desalting Columns (7K MWCO, 0.5 mL, Product #89882).
Target Information
MDM2 is a ubiquitin ligase for p53 and plays a central role in regulating p53 stability. It resides on chromosome 12q.
Akt-mediated phosphorylation of MDM2 at Ser166 and Ser186 enhances its interaction with p300, promoting MDM2-mediated ubiquitination and degradation of p53. Phosphorylation also blocks its binding to p19ARF, further increasing p53 degradation.
MDM2 negatively regulates p53 function by inhibiting its transactivation role. Overexpression of MDM2 can lead to p53 inactivation and diminished tumor suppressor activity.
Additionally, MDM2 possesses E3 ubiquitin ligase activity, directly degrading p53, and plays roles in apoptosis and cell cycle regulation.
MDM2 is overexpressed in many human malignancies, including soft tissue carcinomas and breast cancer. It interacts with proteins such as retinoblastoma 1 and ribosomal protein L5.
More than 40 alternatively spliced transcript variants have been identified in both tumor and normal tissues.
