Target Information

TMP21 (Transmembrane emp24 domain-containing protein 10) is a ubiquitously expressed protein involved in vesicular targeting and protein transport.
It is part of the presenilin complex, modulating γ-secretase but not ε-secretase cleavage activity of the amyloid precursor protein (APP).
The presenilin complex includes APH1, nicastrin, PEN2, and presenilin-1, which together cleave APP at γ- and ε-sites, leading to the formation of the Aβ peptide associated with Alzheimer’s disease.
Co-immunoprecipitation experiments have shown that TMP21 interacts with these proteins, suggesting a regulatory role.
Suppression of TMP21 expression by siRNA increases γ-secretase activity and Aβ production, indicating its modulatory function.
