Target Information

Protein tyrosine phosphatases (PTPs) remove phosphate groups from phosphorylated tyrosine residues on proteins, regulating signaling molecules such as MAP kinases. PTPs are involved in cell signaling, growth, proliferation, and oncogenic transformation. Dual specificity phosphatases (DSPs) selectively dephosphorylate phosphothreonine and phosphotyrosine residues. The VH1 gene in vaccinia virus is a prototypical DSP expressed in late-stage viral infection. VHR’s shallow active site pocket allows hydrolysis of phosphorylated serine, threonine, or tyrosine residues, whereas deeper active sites in typical PTPs restrict substrate specificity to phosphotyrosine.
