Target Information
HMGB1 (High-mobility group box-1) 단백질은 원래 핵 내 비히스톤 DNA 결합 단백질로 알려져 있습니다. 최근 연구에 따르면 손상되거나 괴사된 세포에서 HMGB1이 세포외로 방출되어 염증성 사이토카인으로 작용합니다.
Mouse HMGB1은 215개의 아미노산으로 구성된 단일 사슬 폴리펩타이드로, 두 개의 양전하 DNA 결합 도메인(HMG box A: aa 9–79, box B: aa 89–162)과 음전하의 30 aa C-말단 산성 꼬리 영역을 포함합니다.
잔기 28–44와 180–185는 핵 내 위치 신호(NLS)를 포함하며, B box는 사이토카인 활성을, A box는 전사 인자와의 상호작용을 담당합니다.
HMGB1은 염증 매개체로서 단핵구 이동 및 사이토카인 분비를 촉진하고, T세포-수지상세포 상호작용을 매개합니다.
또한 RAGE, TLR2, TLR4 수용체를 통해 세포 신호를 전달하며, 거의 모든 세포의 핵과 세포질에서 고도로 보존되어 있습니다.
HMGB1은 외상, 패혈증, 허혈-재관류 손상 등 다양한 염증 반응에서 중요한 매개체로 작용합니다.
