Target Information
CD146 (MCAM) is an integral membrane glycoprotein belonging to the immunoglobulin superfamily. It is expressed at cell–cell junctions in endothelial cells and in various normal and malignant cell types. CD146 functions as a calcium-independent cell adhesion molecule and may act as a signal transduction molecule through recruitment of Fyn kinase, leading to tyrosine phosphorylation of intracellular proteins involved in actin cytoskeleton assembly. It plays a role in endothelial cohesion, vascular integrity, and melanoma cell interaction with vascular elements, possibly enhancing tumor spread. CD146 may also function during neural crest cell development and act as a surface receptor triggering phosphorylation of FYN and PTK2 and transient increases in intracellular calcium concentration.
