The Human S100B Simplex ProcartaPlex Kit measures S100B protein and is designed to be combinable with other Simplex kits, allowing users to create customized multiplex panels utilizing Luminex xMAP technology for protein detection and quantitation. When combining multiple Simplex kits, only one buffer kit (sold separately) is required per assay plate regardless of plex size.

ProcartaPlex immunoassays are based on the principles of a sandwich ELISA, using two highly specific antibodies binding to different epitopes of one protein to quantitate up to 80 protein targets simultaneously.

About ProcartaPlex Assays for the Luminex Platform

ProcartaPlex immunoassays employ the Luminex xMAP technology to enable simultaneous detection and quantitation of multiple protein targets. These assays require minimal sample volume (25 μL of plasma or serum, or 50 μL of cell culture supernatant) and deliver analyzed results in approximately four hours.

Technology Overview

ProcartaPlex assays use Luminex beads internally dyed with distinct proportions of red and infrared fluorophores to create spectrally unique signatures identifiable by Luminex detection systems (e.g., Luminex 200, FLEXMAP 3D, MAGPIX). Each bead is conjugated with a capture antibody specific to a single target protein. Detection is achieved through biotinylated antibodies and streptavidin–R-phycoerythrin (RPE), providing quantitative fluorescence signals proportional to protein concentration.

Unlike traditional ELISA, the capture antibody in ProcartaPlex assays is bead-conjugated and free-floating in solution, enhancing multiplexing capability and reducing sample consumption. Luminex instruments use dual lasers to identify bead signatures and quantify RPE fluorescence, enabling simultaneous measurement of up to 80 analytes.

ProcartaPlex multiplex panels are available across six species: human, mouse, rat, nonhuman primate, porcine, and canine.
