Target Information
Tight junction proteins form the cell-to-cell adhesion structures in epithelial cells, comprising the epithelial junctional complex along with adherens junctions and desmosomes. Tight junction strands mainly consist of claudins, occludin, and JAM proteins.
Claudin-16, also known as Paracellin-1, is a member of the Claudin family consisting of 23 integral membrane proteins. It is a renal tight junction protein essential for paracellular magnesium resorption and is localized in the intercellular junctions of epithelial cells in the thick ascending limb of Henle’s loop (TAL marker).
Deficiency or dysfunction of Claudin-16 can lead to impaired renal magnesium handling, potentially causing hypermagnesemia and associated cardiovascular or renal failure. Recent studies indicate that loss of Claudin-16 disrupts paracellular renal transport systems.
