Target Information
Apoptosis(세포자멸사)는 다세포 생물의 정상적인 세포 분화 및 발달 과정에서 발생합니다. TNF(tumor necrosis factor) 및 Fas ligand와 같은 TNF 계열의 사이토카인은 TNFR1(tumor necrosis factor receptor 1)과 Fas 수용체를 통해 세포사멸을 유도합니다.
세포사멸 신호는 death domain(DD)을 포함하는 어댑터 분자 및 ICE(interleukin-1-beta converting enzyme)/CED-3 계열의 프로테아제에 의해 전달됩니다.
새롭게 발견된 DD-함유 분자인 Daxx(death associated protein 6)는 인간, 마우스, 원숭이에서 클로닝되었으며, Fas death domain에 특이적으로 결합하여 Fas 유도성 세포사멸을 강화하고 Jun N-terminal kinase(JNK) 경로를 활성화합니다. Daxx는 인간 및 마우스의 태아 및 성체 조직 전반에서 발현되어 Fas 신호전달 경로에서 중요한 역할을 합니다.
