Anaplastic Lymphoma Receptor Tyrosine Kinase (ALK) belongs to the insulin receptor superfamily and is essential for brain development.
Mutations, rearrangements, and amplifications in the ALK gene have been linked to various tumors, including anaplastic large cell lymphoma, neuroblastoma, and non-small cell lung cancer.
Chromosomal rearrangements often create a fusion gene between ALK (chromosome 2) and nucleophosmin (NPM, chromosome 5), producing the NPM-ALK oncogenic fusion protein.
ALK is a receptor protein-tyrosine kinase with a transmembrane and extracellular domain, showing high sequence similarity to LTK and playing critical roles in neuronal development.
