Product Specific Information
- For Immunohistochemistry (PFA fixed):
Incubate antibody for 30 minutes at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
CD209, commonly known as DC-SIGN, encodes a transmembrane receptor expressed on dendritic cells and macrophages. This receptor plays a key role in the innate immune system by recognizing a wide range of pathogens including parasites and viruses.
It consists of three major domains:
- N-terminal transmembrane domain
- Tandem-repeat neck domain
- C-type lectin carbohydrate recognition domain
The extracellular region functions as both a pathogen recognition receptor and a cell adhesion receptor by binding carbohydrate ligands on microbial and endogenous cell surfaces. The neck region enables homo-oligomerization, enhancing multivalent ligand binding.
Genetic variations in the neck domain can affect ligand-binding ability. CD209 is closely related to L-SIGN (GeneID 10332) but differs in ligand-binding properties and tissue distribution.
Alternative splicing results in multiple variants of this gene.
