Target Information

Spleen tyrosine kinase (SYK) is a cytoplasmic tyrosine kinase involved in immune signaling pathways. Together with ZAP70, SYK is a potential drug target for allergic and autoimmune disorders, and may play a role in cancer.
Upon B cell antigen receptor (BCR) or high-affinity IgE receptor (FcεRI) activation, Syk translocates to the plasma membrane and phosphorylates adaptor proteins, initiating downstream signaling such as calcium mobilization, cytoskeleton remodeling, and transcriptional activation.
Syk interacts with receptor ITAM motifs via its SH2 domains, which are phosphorylated by Src-family kinases that also activate Syk through phosphorylation of its activation loop.
