Target Information
GAD-65와 GAD-67은 group II decarboxylase family에 속하며, L-glutamic acid로부터 GABA(gamma-aminobutyric acid)를 생성하는 반응의 속도 제한 단계를 촉매합니다.
두 단백질 모두 뇌에서 발견되며, GAD-65는 신경 말단의 시냅스 소포막에 국한되고 GAD-67은 세포 전체에 분포합니다.
GAD-67은 기본적인 GABA 합성 수준을 유지하며, 신경전달 과정에서 GABA 수요가 증가할 경우 GAD-65가 일시적으로 활성화되어 GABA 생산을 돕습니다.
GAD-65의 결손은 GABA 신경전달에 장애를 유발할 수 있지만, GAD-67의 결손은 치명적입니다.
대체 스플라이싱으로 인해 GAD-67에는 주요 형태인 GAD-67과 소수 형태인 GAD-25 두 가지 isoform이 존재합니다.
