Target Information
TMP21 is a ubiquitously expressed protein involved in vesicular targeting and protein transport. It is also a component of the presenilin complex and modulates γ-secretase but not ε-secretase cleavage activity of amyloid precursor protein (APP). The presenilin complex includes APH1, nicastrin, PEN2, and presenilin-1. These proteins cleave APP at γ- and ε-sites, potentially leading to accumulation of the Abeta cleavage product associated with Alzheimer’s disease. Co-immunoprecipitation experiments indicate TMP21 may regulate this complex. Suppression of TMP21 expression by siRNA increases γ-secretase activity and Abeta production, suggesting TMP21 modulates γ-secretase function.
