Target Information

CD30 (Ki-1, TNF Receptor Superfamily Member 8) is a type I transmembrane glycoprotein of the TNF receptor superfamily. It was originally identified as a cell surface antigen of Hodgkin’s and Reed-Sternberg cells using monoclonal antibody Ki-1. The ligand for CD30 is CD30L (CD153), and their interaction mediates effects such as cell proliferation, activation, differentiation, and apoptosis.
CD30 plays a critical role in Hodgkin’s disease and other CD30+ lymphomas, acting as a costimulatory molecule in thymic negative selection. It is expressed on Hodgkin’s and Reed-Sternberg cells, certain non-Hodgkin’s lymphomas, virus-infected T and B cells, and activated normal T and B cells.
In T cells, CD30 expression is associated with Th2-type cytokine-producing subsets and CD4+/CD8+ thymocytes co-expressing CD45RO and IL4 receptor. The soluble form (sCD30) serves as a marker for Th2 immune response. TRAF2 and TRAF5 interact with this receptor to activate NF-kappaB signaling. CD30 positively regulates apoptosis and limits autoreactive CD8 effector T cell proliferation, contributing to autoimmunity protection.
Two alternatively spliced transcript variants encoding distinct isoforms have been reported. CD30 is expressed by mononuclear cells in Hodgkin’s lymphoma, Reed-Sternberg cells, and most Anaplastic Large Cell Lymphomas (ALCL), as well as embryonal carcinomas.
