EGFR 유전자는 인간 염색체 7번에 위치하며, HER/ERbB 단백질 패밀리(ERbB2, ERbB3, ERbB4 포함)에 속합니다. EGF(Epidermal Growth Factor)나 TGF-α(Transforming Growth Factor alpha) 등의 리간드가 결합하면 수용체 이합체화 및 세포질 도메인 내 티로신 잔기의 자가 인산화가 유도됩니다.
EGFR 과발현은 두경부, 뇌, 방광, 위, 유방, 폐, 자궁내막, 자궁경부, 외음부, 난소, 식도, 위 및 편평세포암 등 다양한 종양에서 관찰됩니다.
