Product Specific Information

The B-Ly7 monoclonal antibody reacts with human CD103, the alpha E integrin. CD103 non-covalently associates with integrin beta 7 and is primarily expressed on intraepithelial lymphocytes and a subset of peripheral lymphocytes. It is also found in hairy cell leukemia and certain chronic B cell lymphocytic leukemias. Stimulation of human T cells induces upregulation of CD103, which interacts with E-cadherin to mediate lymphocyte homing to the intestinal epithelium.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and validated for flow cytometry using stimulated normal human peripheral blood cells. Recommended usage: 5 µL (0.25 µg)/test in 100 µL final volume.

Super Bright 600 is a tandem dye excited by the violet laser (405 nm) and emits at 600 nm. Recommended filter: 610/20 bandpass. Ensure instrument compatibility.

Staining Buffer Recommendation:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity:
Protect vial and stained samples from light to prevent photo-induced oxidation.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333).

Target Information

CD103 (ITGAE, Integrin alpha E) is expressed on mucosa-associated T lymphocytes and activated cells. It comprises a 150 kDa alpha chain and a 120 kDa beta chain. CD103 is expressed by a small fraction of resting lymphocytes and stains some bone marrow and peripheral blood lymphocytes. As part of the integrin family, it forms heterodimeric membrane proteins that mediate adhesion and activation. Together with integrin beta 7, CD103 binds E-cadherin, forming the mucosal lymphocyte-1 antigen. CD103 dysfunction is associated with hairy cell leukemia and splenic marginal zone lymphoma.
