Product Specific Information

Description:
The SK3 monoclonal antibody reacts with human CD4, a 59-kDa cell surface receptor expressed by thymocytes, mature T helper cells, and low levels on monocytes. CD4 acts as a receptor for HIV, and SK3 blocks HIV binding and mixed lymphocyte reaction. SK3 and RPA-T4 antibodies recognize distinct epitopes.

Applications Reported:
This antibody is suitable for flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use at 5 µL (0.125 µg) per test in 100 µL final volume. Cell number can range from 10⁵ to 10⁸ cells/test.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples may be stored in IC Fixation Buffer (Product # 00-822-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

CD4 antigen is involved in recognition of MHC class II molecules and serves as a co-receptor for HIV. It is primarily expressed in T helper cells but also found in monocytes, macrophages, and dendritic cells. At tissue level, CD4 expression is detected in thymus, lymph nodes, tonsils, spleen, and certain non-lymphoid tissues. CD4 assists in T-cell activation via association with the T-cell receptor complex and Lck kinase, and may mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
