Target Information

Huntington’s disease (HD) is a neurodegenerative disorder characterized by loss of striatal neurons, caused by an expansion of a polyglutamine tract in the HD protein huntingtin. HAP1 was initially identified through two-hybrid library screening; its binding to huntingtin correlates with the polyglutamine tract expansion.

HAP1 interacts with cytoskeletal proteins such as dynactin and pericentriolar autoantigen protein 1, suggesting a role in vesicular trafficking or organelle transport. It also participates in huntingtin-enhanced BDNF transport along cellular microtubules. Attenuation of this process leads to loss of neurotrophic support and neuronal toxicity, potentially contributing to disease pathogenesis. Several alternatively spliced isoforms have been described for HAP1.
