Product Specific Information

Description:
The 16-10A1 monoclonal antibody reacts with mouse CD80 (B7-1), a 55 kDa member of the Ig superfamily. CD80 is expressed by macrophages, dendritic cells, activated B cells, and activated T cells. It binds with high affinity to T cell surface antigens CD28 and CD152 (CTLA-4), playing a crucial role in T-B cell communication and activation.

Applications Reported:
Suitable for flow cytometric analysis.

Applications Tested:
Tested on stimulated mouse splenocytes at ≤0.125 µg/test. Optimal concentration should be titrated empirically (cell number range: 10⁵–10⁸ cells/test).

Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm and is excited at 488 nm (blue laser). Recommended filter: 710/50 bandpass; alternative 695/40 bandpass acceptable. Ensure instrument compatibility.

Light Sensitivity:
Protect vial and stained samples from light due to photo-induced oxidation sensitivity.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using:

• IC Fixation Buffer (Product #00-8222)
• 1-step Fix/Lyse Solution (Product #00-5333)
  Minimal impact on brightness and FRET efficiency.

Excitation / Emission / Laser:
488 nm / 710 nm / Blue Laser

Target Information

CD80 (B7-1) and CD86 (B7-2) are ligands for the T cell costimulatory molecule CD28 and inhibitory receptor CTLA-4 (CD152). Both are expressed on antigen-presenting cells and are essential for T cell activation. CD80 is rapidly induced on activated B cells and various lymphoma cell lines. It provides regulatory signals to T lymphocytes via CD28 and CTLA4 interactions.
Diseases associated with CD80 dysfunction include gallbladder squamous cell carcinoma and myocarditis.
