- Application Notes:
- IHC (frozen): Tissue sections can be fixed in acetone or 2% paraformaldehyde.
- Western blot: Use non-reducing conditions.
- Flow: Weakly stains alveolar macrophages by recognizing the extracellular domain of MARCO; transfected COS cells used as positive control.
- Immunoprecipitation: Immunoprecipitates MARCO as 60 kDa and 50 kDa proteins from transfected COS lysates.
- Functional assays: Blocks human alveolar macrophages by recognizing the extracellular domain of MARCO.

Target Information
MARCO (Macrophage Receptor with Collagenous Structure), also known as SR-A6 or SCARA2, is a macrophage surface receptor and a member of the class A scavenger receptor family. It binds Gram-negative and Gram-positive bacteria via its extracellular, C-terminal scavenger receptor cysteine-rich (SRCR) domain. The protein includes short cytoplasmic and transmembrane domains, an extracellular spacer, and a long collagenous domain that can form trimeric structures. MARCO expression in tumor-infiltrating macrophages and its immunosuppressive role make it a potential target for cancer immunotherapy.
