Target Information

The continued localization of p53 to the nucleus is essential for its tumor suppressor function. PARC, a large Parkin-like ubiquitin ligase, acts as a cytoplasmic anchor protein in p53-associated complexes. Inactivation of PARC results in nuclear localization of p53 and activation of p53-dependent apoptosis, whereas overexpression promotes cytoplasmic sequestration of p53. PARC knockout mice remain viable without obvious phenotype, suggesting redundancy with other E3 ubiquitin ligases such as cullin family proteins. p53 binding to PARC may also regulate PARC function.
