Product Specific Information
- For immunohistochemistry (PFA fixed): incubate antibody for 30 min at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
CD22 (BL-CAM) is a type I integral membrane glycoprotein with a molecular weight of 130–140 kDa. It is expressed in both the cytoplasm and cell membrane of B-lymphocytes. CD22 antigen appears early in B-cell differentiation, similar to CD19. Its membrane expression is limited to mature B cells (CD22⁺) and absent in plasma cells (CD22⁻), making it useful in phenotyping mature leukemias. CD22 is strongly expressed in hairy cell leukemia and is also positive in diffuse large B-cell lymphoma and nodular lymphocyte predominance Hodgkin’s lymphoma, but negative in classical Hodgkin’s lymphoma.
CD22 preferentially binds to α2,6-linked sialic acid. The recognition site can be masked by cis interactions on the same cell surface. Upon ligand-induced tyrosine phosphorylation, CD22 participates in regulation of B-cell antigen receptor signaling, acting as both a positive and inhibitory regulator through interactions with Src family kinases and cytoplasmic phosphatases.
