Target Information

CD22 (BL-CAM) is a type 1 integral membrane glycoprotein with a molecular weight of 130–140 kDa. It is expressed in the cytoplasm and cell membrane of B-lymphocytes and appears early in B-cell differentiation, similar to CD19.
CD22 membrane expression is limited to mature B cells (CD22+) and absent in plasma cells (CD22−), making it useful for phenotyping mature leukemias. It is strongly expressed in hairy cell leukemia, diffuse large B-cell lymphoma, and nodular lymphocyte predominance Hodgkin’s lymphoma, but negative in classical Hodgkin’s lymphoma.
CD22 preferentially binds to α2,6-linked sialic acid, and its recognition site can be masked by cis interactions on the same cell surface. Ligand-induced tyrosine phosphorylation plays a role in regulating B-cell receptor signaling. CD22 functions both as a positive regulator through interaction with Src family tyrosine kinases and as an inhibitory receptor by recruiting cytoplasmic phosphatases via SH2 domains to modulate signal transduction.
