Target Information
The ABL1 proto-oncogene encodes a cytoplasmic and nuclear protein tyrosine kinase involved in cell differentiation, division, adhesion, and stress response.
Activity of c-Abl protein is negatively regulated by its SH3 domain, and deletion of this domain converts c-Abl into an oncogene.
CDC2-mediated phosphorylation regulates its DNA-binding activity, suggesting a role in the cell cycle.
In chronic myelogenous leukemia and certain acute lymphoblastic leukemias, the c-Abl proto-oncogene undergoes a (9;22) chromosomal translocation, producing the Philadelphia chromosome through fusion with the Bcr gene.
c-Abl was initially identified as the viral transforming gene of Abelson murine leukemia virus (A-MuLV) and may regulate DNA repair by activating proapoptotic pathways when DNA damage is irreparable.
