Description:
The 2B8 monoclonal antibody reacts with mouse CD117 (c-Kit receptor), a 145-kDa tyrosine kinase receptor expressed on most hematopoietic progenitor cells, mast cells, and other cell types. The interaction between c-Kit and its ligand (Steel factor) promotes proliferation and differentiation of hematopoietic progenitor cells.
Applications Reported:
Reported for use in flow cytometric analysis.
Applications Tested:
Tested by flow cytometric analysis of mouse bone marrow cells. Recommended ≤0.5 µg per test in 100 µL final volume. Optimize titration for best results.
Fluorochrome Information:
Brilliant Ultra Violet™ 661 (BUV661) emits at 660 nm and is designed for instruments with a 355 nm UV laser. Protect from light due to photo-induced oxidation sensitivity.
Staining Recommendations:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (SB-4401-42) or Brilliant Stain Buffer™ (00-4409-75) to minimize non-specific interactions.
Fixation:
Samples may be stored in IC Fixation Buffer (00-8222-49) or 1-step Fix/Lyse Solution (00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness.

Target Information
KIT (c-KIT):
A proto-oncogene encoding a type 3 transmembrane receptor for mast cell growth factor (stem cell factor). KIT regulates growth and activation of hematopoietic and non-hematopoietic cells. Mutations are linked to gastrointestinal stromal tumors, mast cell disease, and leukemia. c-Kit activation triggers autophosphorylation and downstream signaling through Ras/ERK, PI3K, PLC-γ, and JAK/STAT pathways, essential for hematopoiesis, stem cell maintenance, and gametogenesis.
