Target Information
Histone H2A.X (H2AX)는 히스톤 H2A 패밀리에 속하는 단백질로, 뉴클레오솜 코어 입자를 구성하는 4가지 핵심 히스톤 중 하나입니다.
진핵세포에서 DNA 이중 가닥 절단(DSB)은 H2AX의 C-말단 세린 139의 인산화를 유도하여 γ-H2AX를 형성합니다.
이 인산화는 Western blot 또는 면역형광법으로 검출되어 DSB 빈도를 확인할 수 있습니다.
H2AX 인산화에는 ATM, DNA-PK, ATR 등 인산화 효소가 관여하며, H2A.X는 143개의 아미노산으로 구성되어 있습니다.
Histone H2A.X는 G1과 S기 모두에서 합성되며, mRNA는 polyA tail을 가지거나 가지지 않는 두 가지 형태로 존재합니다.
이 단백질은 인간 염색체 11q23.2–q23.3 영역에 위치하며, DNA 손상 부위에서 MDC1 단백질과의 상호작용을 조절하여 DNA 복구에 중요한 역할을 합니다.
