Product Specific Information

This AD2 monoclonal antibody reacts with human CD73, a 5′-ectonucleotidase that converts 5′-adenosine monophosphate to adenosine. CD73 is expressed on endothelial cells, B and T cells, including some CD4+Foxp3+ regulatory T cells. Adenosine production by these cells has been linked to inhibition of CD4 T cell effector functions such as proliferation and cytokine secretion.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use at 5 µL (0.4 µg) per test.
A test is defined as the amount of antibody that will stain a cell sample in a final volume of 100 µL. Cell number may range from 10⁵ to 10⁸ cells/test.

Compatibility Notice:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for optimal compatibility.

CellBlox Plus Blocking Buffer:
This NovaFluor conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01). Add 5 µL per sample to antibody cocktails prior to combining with cells. For single-color controls, use 5 µL per 100 µL of cell sample.

Target Information

CD73 (Ecto-5′-nucleotidase) catalyzes the conversion of purine 5′-mononucleotides to nucleosides, primarily AMP to adenosine. It forms a dimer of two identical 70 kDa subunits attached to the plasma membrane via a glycosyl phosphatidyl inositol linkage. CD73 serves as a marker of lymphocyte differentiation, and its deficiency is associated with various immunodeficiency diseases. The CD73 gene is localized to chromosome 6q14–q21. Defects can lead to calcification of joints and arteries, and intestinal tuberculosis. Two transcript variants encoding different isoforms have been identified.
