Product Specific Information
This AD2 monoclonal antibody reacts with human CD73, a 5'-ectonucleotidase that converts 5'-adenosine monophosphate to adenosine. CD73 is expressed on endothelial cells and lymphocytes, including CD4+Foxp3+ regulatory T cells. Adenosine production by these cells inhibits CD4 T cell effector functions such as proliferation and cytokine secretion.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells. Use 5 µL (0.4 µg) per test (100 µL final volume). Suitable for 10⁵–10⁸ cells/test.
Compatibility Notes:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
Blocking Buffer Usage:
Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining. Add 5 µL per sample to antibody cocktails or single-color controls (per 100 µL of cell sample).
NovaFluor conjugates use Phiton™ technology based on novel nucleic acid dye structures, engineered for optimized fluorescence performance minimizing spillover and spread.

Target Information
CD73 (Ecto-5'-nucleotidase) catalyzes the conversion of purine 5'-mononucleotides to nucleosides, preferentially AMP to adenosine. It is a dimer of two 70 kDa subunits anchored to the plasma membrane via glycosyl phosphatidylinositol linkage. CD73 serves as a marker for lymphocyte differentiation and is deficient in several immunodeficiency diseases. The CD73 gene is located at 6q14–q21. Mutations can lead to joint and arterial calcification and intestinal tuberculosis. Two transcript variants encoding different isoforms are known.
