Product Specific Information

This AD2 monoclonal antibody reacts with human CD73, a 5′-ectonucleotidase that converts 5′-adenosine monophosphate to adenosine. CD73 is expressed on endothelial cells, B cells, and T cells, including some CD4+Foxp3+ regulatory T cells. Adenosine production by these cells is linked to inhibition of CD4 T cell effector functions such as proliferation and cytokine secretion.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Use at 5 µL (0.4 µg) per test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test.

Compatibility:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.

CellBlox Plus Blocking Buffer:
Updated formulation (Cat. No. C001T06F01) required for optimal staining. Add 5 µL per sample prior to combining with cells. For single-color controls, use 5 µL per 100 µL of cell sample containing 10³–10⁸ cells.

Target Information

CD73 (Ecto-5′-nucleotidase) catalyzes the conversion of purine 5′ mononucleotides to nucleosides, preferentially AMP. It forms a dimer of two identical 70 kDa subunits bound to the plasma membrane via a GPI linkage and serves as a marker of lymphocyte differentiation. CD73 deficiency is associated with various immunodeficiency diseases. The CD73 gene is localized to 6q14–q21. Defects can lead to joint and arterial calcification and intestinal tuberculosis. Two transcript variants encoding different isoforms have been identified.
