Description:
The JES5-16E3 antibody reacts with mouse interleukin-10 (IL-10), an ~18 kDa cytokine also known as Cytokine Synthesis Inhibitory Factor (CSIF). IL-10 is produced by Th2 cells, B1 cells, macrophages, and keratinocytes, and functions to inhibit Th1 cytokine synthesis and T cell proliferation, while providing costimulatory signals for mast cells and Th2 responses.

- Intracellular staining for flow cytometric analysis

Applications Tested:
The Alexa Fluor® 488 conjugated JES5-16E3 antibody has been validated for intracellular immunofluorescent staining with flow cytometry. Recommended usage is ≤ 0.5 µg per 1×10⁶ cells in 100 µL. Titrate for optimal performance.

Target Information
IL-10 encodes a cytokine primarily produced by monocytes and some lymphocytes. It regulates immune responses and inflammation by down-regulating Th1 cytokines, MHC class II antigens, and costimulatory molecules on macrophages. IL-10 enhances B cell survival, proliferation, and antibody production. It can inhibit NF-kappa B activity and is involved in JAK-STAT signaling regulation. Knockout studies in mice indicate IL-10 as a key immunoregulator in the intestinal tract. Mutations in IL10 are linked to increased susceptibility to HIV-1 infection and rheumatoid arthritis. Related diseases include Graft-Versus-Host Disease and HIV-1 infection.
