Target Information
Ataxia-telangiectasia Mutated (ATM) is a protein belonging to the PI3/PI4 kinase family. Mutations in ATM cause ataxia-telangiectasia, a rare autosomal recessive disorder characterized by neurologic degeneration, immunologic deficiency, and increased lymphoid cancer risk.
ATM phosphorylates proteins involved in cell-cycle regulation such as P53, Mdm2, BRCA1, and SMC1, and plays a crucial role in repairing double-stranded DNA breaks induced by ionizing radiation and other mutagens.
ATM autophosphorylation upon DNA damage activates a cascade of kinase reactions regulating cell cycle, apoptosis, and DNA damage repair. It is also linked to apoptosis via Nbs1 and Chk2 in the E2F1 pathway.
