Target Information

PKN2 (PRK2), a close relative of the PKCs, is a regulator of mitosis and exit from cytokinesis. It plays a role in the regulation of cell cycle progression, actin cytoskeleton assembly, cell migration, cell adhesion, tumor cell invasion, and transcription activation signaling processes. PKN2 phosphorylates CTTN in hyaluronan-induced astrocytes, decreasing CTTN ability to associate with filamentous actin. It phosphorylates HDAC5, impairing HDAC5 import. PKN2 acts as a direct RhoA target required for the regulation of the maturation of primordial junctions into apical junction formation in bronchial epithelial cells. It is required for G2/M phase progression and abscission during cytokinesis in an ECT2-dependent manner. Additionally, it stimulates FYN kinase activity necessary for skin cell-cell adhesion during keratinocyte differentiation, regulates epithelial bladder cell migration and invasion, inhibits Akt pro-survival kinase activity, and mediates Rho protein-induced transcriptional activation via c-fos serum response factor (SRF). PKN2 also phosphorylates HCV NS5B, leading to stimulation of HCV RNA replication.
