Target Information

CHEK1 (CHK1) is a serine/threonine protein kinase involved in preventing replication when DNA integrity is compromised.
It is a key mediator in the DNA damage-induced checkpoint network and functions to ensure genomic integrity under genotoxic stress.
Inhibition of CHK1 can abrogate the G2 or S checkpoint, leading p53-deficient cancer cells to mitotic catastrophe and apoptosis, while normal cells remain arrested in G1 phase.
Thus, CHK1 inhibitors can enhance the efficacy of DNA-damaging agents in cancer cells, making Chk1 an attractive therapeutic target, particularly since about 50% of human cancers are p53-deficient.
