Product Specific Information

The DB-1 monoclonal antibody reacts with rat interferon-gamma (IFN-γ), a 15–20 kDa cytokine secreted by Th1 cells.
For optimal detection of rat IFN-γ by sandwich ELISA, use purified DB-1 for capture and biotinylated polyclonal anti-rat IFN-γ for detection.

  • Applications Reported: Capture of rat IFN-γ by ELISA
  • Applications Tested: Tested as the capture antibody in a sandwich ELISA in combination with biotin anti-rat IFN-γ poly (13-7320) for detection and recombinant rat IFN-γ (14-8315) as the standard
  • Recommended Concentration: 1–4 µg/mL for ELISA capture

Target Information

IFN gamma (Interferon gamma, Type II interferon) is a macrophage activation factor produced primarily by T-lymphocytes and natural killer cells in response to antigens, mitogens, and cytokines.
It is a dimeric glycoprotein (~45 kDa) composed of two 146 amino acid subunits. On SDS-PAGE, it appears as 25, 20, and minor 15.5 kDa bands due to differential glycosylation.

Functions of IFN gamma include:

  • Antiviral and tumor antiproliferative activity
  • Induction of MHC class I and II
  • Macrophage activation
  • Enhanced immunoglobulin secretion by B lymphocytes

The biological activity of IFN gamma is highly species-specific; human IFN gamma does not cross-react with mouse IFN gamma.
It acts via IFN gamma receptor I and II, activating the JAK-STAT pathway.
Human IFN gamma shares ~40% sequence homology with mouse IFN gamma and is regulated by IL2, FGF basic, EGF, and downregulated by vitamin D3 or DMN.
Mutations in the IFN gamma gene are associated with aplastic anemia.
