Product Specific Information

The eBioRDR5 monoclonal antibody targets human CD127 (Interleukin-7 Receptor alpha).
CD127 forms a complex with CD132 (common gamma chain) to create the multifunctional IL-7 receptor (IL-7R).
It is expressed on immature B cells, thymocytes, and most mature T cells, with transient down-regulation upon activation.

Functional insights:

• IL-7 binding activates multiple tyrosine kinase pathways, including Jak/STAT.
• IL-7 is crucial for lymphocyte development and T-cell homeostasis.
• CD127 expression is notably down-regulated on CD4+CD25+ regulatory T cells (Tregs), making it a marker for enriched Treg populations expressing Foxp3.

Blocking and validation:

• Binding of eBioRDR5 to PBMCs is blocked by recombinant human IL-7 (Product # 14-1079-80).

Applications Reported:

• Flow cytometric analysis of normal human peripheral blood cells.

Applications Tested:

• Pre-titrated for flow cytometry: 5 µL (0.125 µg) per test (final volume 100 µL).
• Cell count range: 10⁵–10⁸ cells/test.

• Light sensitivity: Protect vial and samples from light.
• Fixation: Compatible with IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333).

It functions as part of the IL-7 receptor complex (alpha/gamma chain heterodimer), mediating signal transduction and cell proliferation in B and T cells.
Associated with immune deficiencies such as severe combined immunodeficiency (SCID).
