Product Specific Information

Antibody detects endogenous levels of TLR5 only when phosphorylated at Ser805.

Target Information

TLR5 is a Type I membrane protein belonging to the Toll-like receptor family, participating in the innate immune response to microbial agents. It mediates detection of bacterial flagellins and acts via MyD88 and TRAF6, leading to NF-κB activation, cytokine secretion, and the inflammatory response. TLR5 binds to TIRAP and MyD88 via their respective TIR domains. It is highly expressed in liver, detected in lung, and at very low levels in most other tissues. The TLR5 gene locus is associated with susceptibility to Salmonella. Mouse strains vary in resistance to S. typhimurium infection, classified as susceptible (BALB/c, C57BL/6, C3H/He), intermediate (DBA/2, C75L), and resistant (A, CBA). The MOLF/Ei strain is highly susceptible, has a unique TLR5 haplotype, and lower TLR5 expression. A common TLR5 stop codon polymorphism that disrupts signaling is associated with susceptibility to Legionnaires’ disease, underscoring TLR5’s role in innate immunity.
