Product Specific Information

This antibody detects endogenous levels of p53 only when phosphorylated at Tyr327.

Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. p53 is up-regulated under stress and activates transcription of genes such as p21waf1, leading to G1 or G2/M arrest. The protein structure includes an N-terminal transactivation domain, a central DNA-binding domain, an oligomerization domain, and a C-terminal regulatory domain. Phosphorylation at various sites, including Ser15 and Ser392, plays key roles in p53 activation and tumor association. p53 functions as a guardian against proliferation of damaged cells and acts as an anticancer factor. Mutations in p53, common in many cancers, disrupt DNA binding and tumor suppressor activity. TP53 gene alterations can occur as somatic or germline mutations, including in Li-Fraumeni syndrome.
