Target Information
CARMA proteins belong to the membrane-associated guanylate kinase-like (MAGUK) family, functioning as molecular scaffolds for signal transduction assembly. CARMA1, CARMA2, and CARMA3 share high sequence and functional homology but differ in tissue-specific distribution, implying distinct biological roles.
The CARD domain of CARMA3 interacts with BCL10, a positive regulator of apoptosis and NF-κB activation. CARMA3 is required for EGF-induced NF-κB activation and contributes to tumor growth in vivo, suggesting its potential as a therapeutic target for EGFR-driven tumors.
