Target Information
Reversible phosphorylation of tyrosine residues is a key regulatory mechanism for numerous cellular events such as proliferation, differentiation, gene expression, and migration. Abnormalities in tyrosine phosphorylation play a role in the pathogenesis of various human diseases, including cancer and immune deficiencies.
There are at least 107 genes coding for protein tyrosine phosphatases (PTPs) in the human genome. All known PTPs contain a conserved 12 amino acid catalytic domain and are categorized based on structural motifs. Receptor-like PTPs have extracellular domains, a transmembrane region, and one or two intracellular phosphatase domains, while nonreceptor cytoplasmic PTPs have a single phosphatase domain with additional motifs.
PTPN5 (STEP), PTPRR, and PTPN7 form a family of phosphatases that inactivate MAPKs (mitogen-activated protein kinases). Multiple STEP isoforms in the adult rat brain are differentially expressed in regions involved in cognitive, affective, and motor functions. These isoforms arise through alternative splicing and have distinct intracellular targets and functions.
