Product Specific Information

The monoclonal antibody OX104 recognizes human CD200 (OX2), a member of the Ig superfamily with two Ig domains, transmembrane, and cytoplasmic regions. CD200 is expressed on resting and activated B cells, subsets of T cells, keratinocytes, peripheral and central nerve cells, follicular dendritic cells, and ovarian cells. Interaction with CD200R regulates macrophage activation and reduces inflammatory cytokine secretion.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Tested on normal human peripheral blood cells at ≤5 µL (1.0 µg)/test. Recommended careful titration for optimal performance.

Note:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL of CellBlox Plus per sample or per 100 µL of cell sample for single-color controls.

Target Information

CD200 is a type-1 membrane glycoprotein with two immunoglobulin domains, belonging to the Ig superfamily. It interacts with CD200R on myeloid cells to inhibit macrophage activation. Homologues are found in herpesviruses and poxviruses. CD200 may regulate myeloid cell activity and deliver inhibitory signals to macrophages. Dysregulation of CD200 is associated with Mantle Cell Lymphoma and Primary Hypertrophic Osteoarthropathy.
