Product Specific Information

The 236A/E7 antibody reacts with human Foxp3 (FORKHEAD BOX P3, SCURFIN, JM2), a 49–55 kDa transcriptional regulator. Foxp3 is highly expressed in CD4+CD25+ regulatory T cells (Treg) and can induce a Treg phenotype when ectopically expressed in CD4+CD25− cells.

Intracellular staining and flow cytometric analysis of human PBMCs using this antibody and the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523) show staining of the CD4+CD25^bright population.

The 236A/E7 epitope is distinct from that of clone PCH101. This antibody also recognizes rhesus macaque, sooty mangabey, and cynomolgus macaque.

Applications Reported: Intracellular staining and flow cytometric analysis.
Applications Tested: Pre-titrated and validated with the Foxp3/Transcription Factor Staining Buffer Set. Recommended usage: 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorochrome Information:
eFluor® 660 replaces Alexa Fluor® 647, emits at 659 nm, and is excited by a 633 nm red laser.
Excitation: 633–647 nm; Emission: 668 nm; Laser: Red.
Filtered post-manufacture (0.2 µm).


Target Information

FOXP3 (Forkhead box protein 3) is a transcriptional regulator essential for immune homeostasis. It is expressed at high levels in CD25+CD4+ regulatory T cells, low levels in CD4+CD25− cells, and absent in CD4−CD8+ T cells. FOXP3 acts as a master regulatory gene and marker for Tregs. Mutations cause the scurfy phenotype in mice and IPEX syndrome (immune dysfunction, polyendocrinopathy, enteropathy, X-linked) in humans. FOXP3 expression in human breast and colon cancer cells is regulated by p53 in response to DNA damage.
