Target Information
CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase (PTP) expressed in all nucleated hematopoietic cells, accounting for approximately 10% of lymphocyte surface proteins. It is absent in non-hematopoietic tissues and cell lines.
CD45 plays a crucial role in lymphocyte development and antigen signaling by regulating Src-family kinases. Multiple isoforms exist due to alternative splicing, differing in extracellular domains but sharing transmembrane and cytoplasmic regions.
CD45RA, one isoform, shows restricted expression among lymphoid cell subtypes. Isoforms vary in membrane localization and expression depending on cell type and physiological state.
CD45 regulates T- and B-cell receptor signaling, suppresses JAK kinases for cytokine signaling, and promotes cell survival through integrin-mediated pathways, apoptosis regulation, and modulation of immune functions.
