Terminal deoxynucleotidyl transferase (TdT) is a unique deoxynucleotide polymerizing enzyme (~58 kDa) found in B- and T-cell lymphoblasts/prelymphocytes. TdT contributes to antigen receptor diversity by synthesizing non-germline elements (N-regions) at the junctions of rearranged Ig heavy chain and T-cell receptor gene segments.
Rare TdT-positive cells are regularly detected in thymus and bone marrow. Expression varies significantly among thymic cells and decreases in mature T-cells. Occasional TdT-positive cells can be found in tonsils, lymph nodes, and extranodal lymphoid tissues.
Immunohistochemical detection of TdT is valuable in classifying malignant lymphomas and acute leukemias, particularly for identifying pre-B and pre-T acute lymphoblastic leukemia/lymphoblastic lymphoma (ALL/LBL).
Target Information
Terminal Deoxynucleotidyl Transferase (TdT) is a nuclear DNA polymerase that catalyzes the polymerization of deoxynucleotides at the 3′ hydroxyl ends of oligo- or polydeoxynucleotide initiators, functioning without a template.
TdT is a highly specific marker for diagnosing and classifying acute lymphoblastic lymphoma/leukemias and is valuable for differentiating lymphoblastic lymphoma from Burkitt’s lymphoma.
