Specificity Comments:
Cytoskeletal intermediate filaments (IFs) are tissue-specific proteins used as markers to distinguish cell types and identify tumor origins. Desmin, a class III IF protein (469 amino acids) located near the Z line in sarcomeres, is expressed in adult differentiated muscle tissues. Anti-desmin detects cells of normal smooth, skeletal, and cardiac muscles. The antibody reacts with leiomyomas, leiomyosarcoma, rhabdomyomas, rhabdomyosarcoma, and perivascular cells of glomus tumors of the skin.

Target Information

Desmin (~53 kDa) shows high tissue specificity, being expressed predominantly in muscle cells (cardiac, skeletal, and smooth). Its expression is stage- and tissue-specific, induced during terminal differentiation of skeletal muscle cells. In skeletal and cardiac muscle, desmin localizes to the Z-disk and intercalated disk. Expression in smooth muscle is more heterogeneous. Coexpression of vimentin and desmin is observed in muscle-derived tumors such as rhabdomyosarcomas and leiomyosarcomas. Altered desmin distribution has been noted during myocardial dysfunction.
